{Abstract}: The Pleurotus pulmonarius LAU 09 was characterized to the species level and registered at the GenBank database (accession number: JF736658). P. pulmonarius LAU 09 formed a phylogenetic cluster with P. pulmonarius complex with bootstrap value of 77%, which confirmed the close evolutionary lineage with previous characterized P. pulmonarius complex. The polysaccharide obtained from P. pulmonarius was structurally characterized and tested for antimicrobial and anti-inflammatory activities. The polysaccharide was active against all tested pathogens except Pseudomonas aeruginosa with percentage activity of 85.75%. The highest zone of inhibition was obtained against Staphylococcus aureus (30 mm), while the lowest zone size obtained against Escherichia coli (7 mm). The polysaccharide revealed the potent anti-inflammatory effects against carrageenan and formalin - induced paw edema in rats with higher inhibitory percentage in rats writhing; 83.33% and 92.37% inhibition were obtained in carrageenan and formalin-induced paw edema in rats treated with polysaccharide compared with 33.33 and 84.73% in rats treated with standard (dichlofanac) respectively. The percentage sperm motility were higher in both carrageenan and formalin-induced paw edema in rats treated with polysaccharide (76.25 and 67.95%) while its low in dichlofenac treated rats (47.35 and 63.20%) respectively. The sperm counts were increased considerably in the rats treated with polysaccharide (carrageenan: 44.75 x 10(6), formalin: 44.80 x 10(6)) compared with the control, 24.70 x 10(6) (carrageenan) and 42.00 x 10(6) (formalin). The significant (p < 0.05) increased obtained in both carrageenan and formalin-induced paw edema in rats treated with polysaccharide in some blood parameters in comparison with dichlofenac treated rats was an evidence of anti-inflammatory potential of the polysaccharide.

{Abstract}: The mycelia of Tricholoma Mongolicum M-1 was used to study the effects of various carbon and nitrogen sources, initial pH, temperature, inoculums size, rotation speed and medium capacity on the production of extracellular polysaccharide (EPS) and intracellular polysaccharide (IPS) in shake-flask cultures. The optimization of submerged culture conditions were studied using single factor test and orthogonal matrix method. The results showed that the optimal medium contained 30 g/L yeast extract, 20 g/L glucose, 0.3% KH2PO4 and 0.3% MgSO4, then the optimal cultivation conditions were: pH 6.5, temperature 25 degrees C, inoculums size 12% and rotation speed 180 rpm. Under these optimal conditions, the maximum EPS and IPS concentrations reached 1.187 and 1.073 g/L respectively, which were twice as much as the yields at the basal medium.

{Title}: Effect of the extracts from Gastrodia elata BL. on mycelial growth and polysaccharide biosynthesis by Grifola frondosa

{Abstract}: Two groups of the extracts from different Gastrodia elata BL.s were separately aadded into the fermentation system of Grifola frondosa in submerged culture. The results showed that the two extracts could promote the biomass growth of Grifola frondosa significantly (P <0.05). However, the extracts of Gastrodia elata BL. in the group 2 would more contribute to Grifola frondosa's biomass growth than that in the group 1, and the extracts of Gastrodia elata BL. in the group 2 containing 5% (v/v) markedly promoted the biomass and extracellular polysaccharide (EPS) biosynthesis of Grifola frondosa (P<0.05). Biomass and EPS production increased from 0.564 +/- 0.09 to 1.324 +/- 0.25 g/l and from 71.69 +/- 0.53 to 107.08 +/- 0.85 mg/l, which increased by 134.75 and 49.37%, respectively. However, intracellular polysaccharides (IPS) content declined from 60.38 +/- 0.87 to 45.71 +/- 0.66 mg/g, which decreased by 24.30% compared with the control's, respectively. Moreover, a fact showed that EPS-2 and IPS-2 were both the main components, and there were no new other polysaccharide components separated by DEAE-52 column in the sample. It suggested that the polysaccharide biosynthesis pathways of Grifola frondosa may be unchanged.

{Abstract}: The extraction parameters for Pholiota adiposa SX-01 exopolysaccharide (EPS) produced during submerged culture were optimized using response surface methodology. The optimum conditions for EPS extraction were predicted to be, concentration at 87.36 degrees C, precipitation for 35.18 h at pH 8.81, and EPS production was estimated to be 8.45 g/L. The actual yield of EPS under these conditions was 8.39 g/L. The in vitro antioxidant activities of the EPS were investigated and results showed that the inhibition effects of EPS at a dosage of 250 mg/L on superoxide anion radical, hydroxyl radical and reducing power were 70.63 +/- 5.26, 58.22 +/- 5.41 and 0.37 +/- 0.02% (absorbance at 700 nm), respectively. The results provide a reference for large-scale extraction of EPS by P. adiopsa SX-01 in industrial fermentation.
