{Abstract}: Tolypocladium sp. Ts-1 was isolated from the fruiting body of a wild Cordyceps sinensis, one of the best known traditional Chinese medicine and health foods. The antioxidant activities of hot-water extracts from cultured mycelia of Tolypocladium sp. were assessed in different in vitro systems. The extracts showed superoxide dismutase (SOD) activity of 35.6 U/mg protein and are effective in scavenging superoxide radical in a concentration dependent fashion with IC50 value of 1.3 mg/mL. As a reinforcement of the action, similar radical scavenging effects of the extracts were also discerned with both site-specific and non site-specific hydroxyl radical using the deoxyribose assay method. 1,1-Diphenyl- 2-picrylhydracyl (DPPH) radical scavenging activities of the extracts reached more than 75.2% at the concentrations of 3-6 mg/mL. The extracts showed moderate reducing power and ferrous ion chelating activity. Moreover, the protective effects of the extracts against hydrogen peroxide (H2O2)-induced lesion to rat PC12 cell (pheochromocytoma cell line) was observed in a dose-dependent manner from 0.25 to 2.0 mg/mL. Our results suggest that the aqueous extract of Tolypocladium sp. mycelium has strong antioxidant activities and is a potential source of natural antioxidant products. This is the first report on antioxidant activity of Tolypocladium fungus isolated from wild C. sinensis, an endangered species.

{Abstract}: The extraction parameters for Pholiota adiposa SX-01 exopolysaccharide (EPS) produced during submerged culture were optimized using response surface methodology. The optimum conditions for EPS extraction were predicted to be, concentration at 87.36 degrees C, precipitation for 35.18 h at pH 8.81, and EPS production was estimated to be 8.45 g/L. The actual yield of EPS under these conditions was 8.39 g/L. The in vitro antioxidant activities of the EPS were investigated and results showed that the inhibition effects of EPS at a dosage of 250 mg/L on superoxide anion radical, hydroxyl radical and reducing power were 70.63 +/- 5.26, 58.22 +/- 5.41 and 0.37 +/- 0.02% (absorbance at 700 nm), respectively. The results provide a reference for large-scale extraction of EPS by P. adiopsa SX-01 in industrial fermentation.

{Abstract}: The polysaccharide fraction, CM-hs-CPS2, was isolated from fruiting bodies of cultured Cordyceps militaris grown on solid rice medium by hot water extraction and ethanol precipitation, and purified by ion exchange column (DEAE-cellulose-52) and gel filtration column (Sephadex G-100) chromatography. Its structural characterizations were analyzed by gas chromatograph (GC) and fourier transform infrared spectroscopy (FT-IR). Results indicated that CM-hs-CPS2 was a kind of sulfating acidic polysaccharide containing acetamido group. The monosaccharide composition analysis showed that CM-hs-CPS2 was composed mainly of mannose, glucose and galactose in a molar ration of 1.35: 8.34: 1.00, and linked by a-glycosidic linkage. The studies on antioxidant activities of CM-hs-CPS2 were evaluated by various methods in vitro. Results showed that DPPH. scavenging activity, reducing power and ferrous ion chelating activity of CM-hs-CPS2 (8 mg/ml) were 89%, 1.188 and 85%, respectively.

{Abstract}: Black soybeans [Glycine max (L.) Merrill] were fermented with various GRAS (generally recognized as safe) microorganisms, including Aspergillus sp., Bacillus sp. and yeast (Issatchenkia orientalis) at 30°C for 3 d. The antioxidant contents, including total phenolics and total flavonoids, and the antioxidant activities of 1,1-diphenyl-2-picrylhydrazine (DPPH), ferric reducing antioxidant power (FRAP) and HO.scavenging were analyzed in preparations of fermented black soybeans. Fermented cultures proved to yield significant levels of antioxidants compared with non-fermented cultures (P < 0.05). Fermented black soybean preparations possessed antioxidant activities could be attributed to the total phenolics and flavonoids present.
